DNase I 是一種具DNA特異性的內切酶，可將 dsDNA 或 ssDNA 水解為寡核苷酸和單核苷酸的混合物。可用於RNA萃取、體外轉錄 (in vitro transcription)、RT-PCR 中去除 DNA、DNase I 足跡分析實驗 (DNase I footprinting)、nick translation、DNA library 製備等分子生物學實驗。

DNase I 全名為 Deoxyribonuclease I，是一種能夠消化單股或雙股 DNA 的核酸內切酶 (endonuclease)，能辨別並切割磷酸二酯鍵 (phosphodiester bond)，產生單個脫氧核苷酸或具有 5'-磷酸化 (5'-phosphorylated) 和 3'-羥基化末端 (3'-hydroxylated end) 的單鍊或雙鏈寡核苷酸 (oligodeoxynucleotide)。

DNase I 的活性依賴於 Ca2+，並可被二價金屬離子 Mg2+、Mn2+ 等活化。在 Mg2+ 存在的情況下，DNase I 可隨機識別並切割雙鏈DNA 任意一條鏈上的任意位點；而在Mn2+ 存在的情況下，可識別並切割DNA兩條鏈上幾乎相同的位點，產生平末端 (blunt end) 或有1-2 個核苷酸突出的粘性末端 (sticky end) 的DNA片段。
