Bucculite™ FdU Cu Free Cell Proliferation Fluorescence Imaging Kit 細胞增生螢光檢測套組

測定新合成的 DNA 是偵測增殖細胞最可靠、最精確的方法。其中傳統的方法是於細胞週期的 S 期將胸腺嘧啶類似物 (thymidine analog) - BrdU (Bromo-deoxyuridine) 併入新複製的 DNA 中，後續可使用螢光標記的抗 BrdU 抗體來偵測 BrdU 標記的增殖細胞，但 BrdU 法的步驟繁多，需要使用BrdU抗體，影響因素較多且穩定性比較差。而 AAT Bioquest 提供 Bucculite™ FdU Cu Free Cell Proliferation Fluorescence Imaging Kit 使用一種代替抗 BrdU 抗體和 EdU 的點擊化學 (Click chemistry) 檢測法，不需使用抗體、操作便捷、檢測靈敏度高，是一種環境友善的無銅檢測方法，可於細胞中檢測活性的 DNA 合成。

Bucculite™ FdU Cu Free Cell Proliferation Fluorescence Imaging Kit 使用胸腺嘧啶核苷的類似物 FOL-FdU (Fluorodeoxyuridine) 取代 BrdU 來標定 DNA。因 FOL-FdU 在 DNA 合成過程中會被併入細胞的 DNA 中，細胞固定後，接著使用螢光結合的 MTA-iFluor® 來標定併入的 FOL-FdU，FOL 會與 MTA 產生交聯反應，且無須任何的催化劑。最後可於相對應的螢光濾片組中，觀察細胞中 iFluor® 螢光標定的 DNA。
