Blocking reagent - Normal Serum 動物正常血清

因 BSA 與奶粉中含有許多牛源的蛋白，做為遮蓋/封閉劑 (blocking agent) 時可能會與 bovine、goat、sheep 等來源之抗體交互作用而產生背景雜訊，若以與二抗同物種之 normal serum 做為遮蓋/封閉劑，可有效降低非專一性訊號。

動物正常血清取自於未免疫的動物，經過脂質萃取以提高透明度，再以 PBS 進行透析，最後冷凍乾燥保存。正常血清可提供足量之內生性蛋白用以飽和 (Saturate) 及遮蓋/封閉 (Blocking) 待測樣品上的非特異性結合點。在免疫組織染色 (IHC) 或免疫螢光染色 (IF) 時遮蓋/封閉 (Blocking) 使用時，一般為濃度 5-10% (v/v) 稀釋於 PBS 緩衝液中。
需注意的是使用間接 (indirect) 檢測時，血清應與帶標記 (螢光或酵素) 之二抗的宿主來自同一物種，如此即可降低背景值。
